HLA Typing

Unique Aspects of Laboratory Testing

  • High resolution types are resolved to a SINGLE allele.
  • Homozygous types are confirmed using at least two typing methods, family pedigree or linkage analysis.
  • Rare alleles and unusual combinations of alleles are confirmed.
  • Novel alleles are fully characterized using selective amplification or cloning.
  • High resolution typing is performed using automated nucleotide sequencing supplemented with SSP and SSO methods as required to resolve ALL ambiguities.
  • Expert consultation is available for all donor searches and selection.

Single Allele Resolution

Tests

High resolution typing is available for the following loci:
HLA-A HLA-DRB1 HLA-DPB1
HLA-B HLA-DRB3/4/5 HLA-DQA1
HLA-C HLA-DQB1 HLA-DPA1

Reduced cost for packages:
A, B, C, DRB1 and DQB1
A, B, C, DRB1, DQB1 and DPB1
A, B, C and DRB1

Indications

  • To determine HLA matching between transplant recipient and
    donor(s)
  • To confirm HLA identity for related individuals
  • To identify HLA types required for immunotherapy
  • To diagnose certain HLA-associated diseases

Methods

  • Sequencing
  • SSO, SSP and/or Cloning to resolve ambiguities

Turn Around Time (arrival before 10am)

Routine test results in 4-10 business days

STAT testing in 2-3 business days

Low/Intermediate Resolution

Tests

Low/Intermediate resolution typing is available for the following loci:
HLA-A HLA-DRB1
HLA-B HLA-DQB1
HLA-C HLA-B27

Reduced cost for packages:
A, B and C
DRB1 and DQB1

Indications

  • To determine HLA types of family members
  • To screen unrelated donors for bone marrow transplantation
  • To evaluate donors for solid organ transplants
  • To diagnose certain HLA-associated diseases

 Methods

  • SSO and SSP

Turn Around Time (arrival before 10am)

Routine test results in 2-5 business days

STAT testing in 1-2 business days

Serology

Tests

HLA-A, -B and -C
HLA-AB Expression (confirms expression of DNA-based HLA types determined by DNA typing performed by this lab)

Indications

Confirming expression of types determined by DNA-based method

Methods

  • Microcytotoxicity

Turn Around Time (arrival before 10am)

Routine test results in 2-4 business days

STAT testing in 1 business day
